Can I run a subs and an amp to my stock radio?

When you say "the wires in your truck", do you mean the wires that are running to the stock amp in the trunk that came with the car right?
Also, is it possible to just run subs directly to current amp? That way I could avoid the whole aftermarket amp and line out connector, or do I need to run an aftermarket amp?

If you are unfamiliar with wiring and have to Google everything. Should take about an hour or two. I did mine in 15 mins, but electronics are my job.

What size sub do you have? I have the option of buying one or two 12 inch ones. Is one enough bass?

It depends on what you mean by "enough" and what the RMS ration of the subs is. Also what the RMS of the amp is. I have two 12" 4ohm subs rated at 1500watts RMS, being pushed by a dual channel amp rattled at 2000w RMS @ 2 ohms. Im getting more than enough sound to shake to block if need be so it all just depends on what you want.
